How do I integrate my thunderbird as email client for Pepipost or other SMTP servers?
I want to add SMTP server Pepipost to let me send email/newsletter to our members. How do I setup the SMTP server so that I can send mail. How is this relevant to adding an incoming server?

That does not answer my question, I have gone through all the links and descriptions but nothing seem to work. I have integrated my domain with pepipost. Now how do I start sending mails. Do I need to setup a mail server or so before sending the mail from email client or what?
After you add the pepipost smtp, select the account from which you send mail, in the left pane of Account Settings, then click the Outgoing Server (SMTP) drop-down in the lower right pane and select the pepipost smtp.
Its already done, what's next. How do I start sending mail with no email address present in it? I only have my domain integrated with pepipost.
You will have to add a mail account to TB and then point it to the pepipost smtp. If you've already registered your domain with pepipost, create a mail account with your domain host and add that, using the settings provided by the host.
Then when I send mail from the mail account em1, which address would it be sent from. Also if the receiver responds with a reply, where will it land. This is complicated. You mean I have to add a mail account (gmail, hotmail) to it to receive mails. But how do they point to my email address. How can I make multiple username aliases.
If you send from em1, the recipient will reply to em1 or the Reply-To address for em1. That is, if pepipost allows the em1 provider to send through the pepipost smtp. That is the purpose of registering your domain with pepipost: it allows a domain mail account to send on the pepipost smtp. I don't think that would work with any hotmail or gmail account, otherwise every spammer would send on the non-secure pepipost smtp.
The TB element of your question has been answered - direct questions about domain mail accounts to your domain host and pepipost.
How do I get the domain mail account? Is it any different than gmail or gsuite? How is mailtrain relevant in this topic?

If you already have abc@domain.com etc. accounts created and working, add them to TB from File/New/Existing Mail Account, using the settings provided by the domain host. Once they are in TB, set them to send through the pepipost smtp. You can't just enter an address in TB that hasn't been created with a mail service.
Is pepipost not a mail service (any other example. then what is TB?) apart from a SMTP server and yes I have no abc@domain.com account but only the domain. Then what was the integration then with pepi, if we cannot generate user names or aliases to me correct.
I send abc to my members, def to my support people, ghi to accountants and so on if it is possible to variate the username but keep the domain name same.
Pepipost is a service to send commercial-level numbers of messages that wouldn't be allowed by a standard mail service. But you must have a mail account with the domain that is registered with pepipost. Creating accounts with the domain is not a TB issue, but once you create the accounts they can be added to and accessed by TB.
But get this, pepipost support says there is no need of a mail account with the domain, they keep insisting. Also the link you sent foremost did not contain any mail service indications. Thanks sfhowes for your support. I hope am not bothering you with much.

Since iskconjaipur.com is hosted on net4india.com, I suggest you contact them for the POP/IMAP/SMTP settings to add the account to TB.
I think I have found the answer. Just needed to change the default identity in Account settings for my gmail to pepi one and select the appropriate outgoing SMTP server. Then it asked for the password. Voila. Now just have to figure a way to use multiple email address like "memo@iskconjaipur.com" with other instances such as "member@iskconjaipur.com", "support@iskconjaipur.com" etc. Any suggestions.

I would have done it a bit differently, i.e. keep the gmail address in the Email Address box, and select the gmail smtp as the Outgoing Server. This would keep the gmail account separate from the others and fully functional. Then, click Manage Identities and add memo@isk...com in the Email Address, isk...@gmail.com as the Reply-to Address, and the pepipost smtp server in Outgoing Server. I assume the existing pepipost smtp has memo@isk... as the User Name, but if you were to add other identities such as support@isk..., you might have to create another pepipost smtp with support@isk... as the User Name. Give each smtp a distinct Description, e.g. pepisupport, pepimemo etc., to make it easy to match the identity with the right smtp.
When you write a new message, click the From: drop-down to select the desired identity. At the moment, replies to all mail sent from @isk... addresses are sent to the gmail account, but to have all mail that is sent to @isk... addresses be directed to the gmail account, you would have to open the settings on the gmail website and have gmail 'fetch' the @isk.. accounts. Or, add each @isk... address to TB as a separate account .
In the Copies & Folders tab of the New Identity window, you may wish to have sent mail copied to a folder named after the identity.

Would you suggest how can I automate this if I have users from our website subscribe for newsletters or want to receive notifications about the daily happenings in my organisation (am IT volunteer at an NGO) to send mails automatically.

- As far as fetching using gmail for the @isk.. address, these address are not actually existing on the SMTP server or my domain, they are just username aliases for sending mail but not for receiving. Don't know how they work with SMTP to receive back. I guess that is where other service may come in (which I dont know about).

To send newsletters or notifications to groups, consider mailing lists. To automate mailings, filters are an option, or perhaps create an RSS newsfeed for users to subscribe.
