What is the best way to backup all emails including local folders?

I'm not sure how Thunderbird stores the email messages or where they are on the hard drive. I use Norton 360 to backup my hard drive, but I'm not sure if it gets the Thunderbird emails or just files from my old Windows Live Mail.

I also have several local Thunderbird folders I would like to backup.

Does TBird profile local store directory also contain all local folders, or can I have separate local folders in different local directories? I use separate encrypted "drives" to house confidential client email files, thus need 1 separate email local store file per client. Is this possible in TBird?
