I have some security-themed extensions (like BlockSite). How do you prevent someone from going into the add-on manager and simply disabling or uninstalling an extension in order to bypass it (like preventing my kids from disabling BlockSite in order to bypass its functions)? Using FF 4.01.

You could use another extension, Public Fox, to password protect certain parts of Firefox including the add-ons manager - https://addons.mozilla.org/firefox/addon/public-fox/

As with using any extensions used to protect Firefox, they can easily be bypassed by creating a new profile.
