I would like to set up my inbox so that the unread messages are always at the top and then have the read messages sorted so that the starred messages are above the non-starred ones. I can sort by read status to get the unread messages at the top, but don't see a way to sort the read messages after that. Is there a way to do this?

If you sort the list by read/unread, you can't then sort just the read by star, but if you had a custom view to show just read mail, clicking the Starred column would sort into starred and non-starred groups.

To make a custom view, right-click the Mail Toolbar, Customize, drag the Views icon onto the toolbar, then create a new view by clicking the View drop-down, Customize, and add a view with Status + is + Read.
