I only have one website login saved whereby I want Firefox to automatically fill-in when I visit it. The issue is that when I uncheck "Ask to save logins and passwords for websites" even Saved Logins no longer automatically get filled-in!

Yes, that checkbox is a "master switch" for the whole Password Manager feature. I don't know if there is a workaround, other than turning it on when you need it and then off again. Not very convenient, I know.

You should still be able to use "Fill Login" and "Fill Password" in the right-click context menu, but this might not be easy on Mac if you do not have a two button mouse.

An alternative might be to use Firefox Lockwise.

That is a shortcoming of the feature and a poorly written control given "Ask to save logins and passwords for websites" explicitly deals with the saving of passwords.
