If I open Bookmarks and right-click on a website, why are "Open bookmark in a new Tab" and "Open Bookmark in a new window" greyed out? In previous versions, I could just open a bookmarked item in a new tab, but Firefox 4 won't allow me to do this.

You might be able to achieve your goal by installing this add-on: Open Link in New Tab

Thanks for your response. It's very strange - the problem seems to have corrected itself, and I haven't run into it since.

Sometimes I think Firefox has a mind of its own ;)

Firefox doesn't have such context menu entries for "Open Bookmark ...". So it looks that there has been a problem with the extension that added them and that started working again.
