Your profile has your mail, so can you go into the Recycle Bin and restore the folder?

If you do not need that folder back, you can create a new profile.

Pop up the Windows "Run" dialog by holding down the Windows key on the keyboard and tapping the R key.

Paste this command line and then click Run to execute it:

"C:\Program Files\Mozilla Thunderbird\thunderbird.exe" -P

Don't delete anything you see here (even if it's dysfunctional, in case you want to restore the old folder later) just create the new profile and start TB in the new profile.
