I'm finding videos on certain websites stop responding to mouse clicks, or the spacebar key (play/pause shortcut), soon after opening full-screen mode.

I ran the SEO Site Checkup flash test for one such site and it reported no Flash being used on the site. (https://seositecheckup.com/tools/flash-test)

Seems to be no problem when playing YouTube videos.

Not able to reproduce the problem in Google Chrome.

Just to add that at the time the video stops responding it may be paused, or playing, so will remain paused, or playing. Similarly the Esc key shortcut still works to exit full screen mode and the other actions/controls items on the website behave as normal (scrolling, buttons).

Furthermore the website in which I've just seen this issue also has other videos (tesimoials) below the main video, which don't exhibit the same issue. They appear to be YouTube or Vimeo videos and pop up in a player window, unlike the main video.

If I open the website in a new tab, it responds normally (unless I go to full screen mode)

Closing all running instances and restarting Firefox, clearing site data, cookies and history doesn't resolve it. After exiting full screen mode, and refreshing, the video behaves normally again, unless you go into full screen mode.
